    The RAPID system is a field-hardened rapid thermal cycler with fluorescence monitoring and capable of automatically analyzing samples for the presence of any nucleic acid sequence. An advanced artificial intelligence (IE) system allow the RAPID to automatically collect the data, interpret the data, report the results and provide real-time reach-back capability through a secure Internet connection.

    The RAPID system includes an IBM Thinkpad laptop computer, backpack, bubble centrifuge, start-up kit, basic tool kit, and user manual. At 50 pounds the system is lightweight, portable and is air, land and sea transportable.

    The RAPID is a field-hardened rapid thermocycler with concurrent fluorescence monitoring. The RAPID is capable of automatically analyzing samples for the presence of any DNA sequence. Easy-to-use software allows the RAPID to automatically collect the data, interpret the test data, and report the results in less than 30 minutes..

    FAST: Up to 32 prepared test samples may be amplified and analyzed in less than 30 minutes.

    Easy to use: The software is divided into basic and advanced modes. Analysis protocols are designed and tested by lab personnel in advanced mode, and then established as a push-button instruction. Field personnel can then prepare samples, place them in the instrument, and push one button. The RAPID then runs the appropriate reaction, analyzes the florescence change in the samples, and displays the results in an easy to understand format. Test results can be printed on commercially available printers.

    Portable: The RAPID is designed to be air, land, and sea transportable; able to withstand unrestrained collisions with the bed and sides of military and commercial transport vehicles in packaged configurations and operate after exposure to loose cargo vibrations while in transport mode. The instrument and all accessories weigh a total of 50 lbs., and may be manually transported with the included backpack.

    Rugged: The RAPID was built for shock resistance. It is capable of withstanding a 1 meter drop in transport mode, and a 15 cm drop while operating. The RAPID can be transported under a variety of environmental conditions including smoke, dust, rain, salt spray, high humidity, temperature extremes, reduced atmospheric pressure, and sand.

    The RAPID is based on LightCycler® 32 Instrument Technology. Like the LightCycler Instrument, the RAPID is a high-speed thermocycler with a built-in fluorescence monitoring system. Up to 32, 5 to 20 ul samples are centrifuged into the tips of composite glass/plastic reaction vessels. These are placed in a rotating carousel, which positions the samples at the focus of fluorimeter optics in a cylindrical reaction chamber. A central fan spins at 2,000 - 10,000 rpm creating a turbulent vortex of air inside the reaction chamber. This ensures highly uniform temperatures and rapid temperature transitions. Heating and cooling are achieved by varying fan speed and a heating coil. Interchangeable optics modules allow fluorescence excitation and monitoring over a wide range of wavelengths.

    Reactions can be monitored using Hybridization Probes and double-strand DNA specific dyes, such as SYBR Green 1*. This allows monitoring cycle-by-cycle accumulation of amplification products and coupled with a novel approach to product differentiation, allows quantification of low initial template copy numbers.

    The RAPID was designed to take the unique analytical capabilities of the LightCycler Instrument to the field. Throughout the design process, the contradictory needs of decreased weight and increased durability had to be carefully balanced in order to achieve a workable final design. The system had to be portable, yet tough enough to withstand a 1 meter drop in transport mode without instrument damage, and a 15 cm drop during operation without interrupted functionality. In the redesign process, some of the areas of concentration were reducing the impact susceptibility of the optics system, strengthening the electronic circuitry, integration of the larger 1.5 mm tube design for a larger sample size and decreased sample fragility, an integrated transport case, and a backpack system to allow portability of the instrument.
